What might work is just batch all of the TIFF files through an image converter to an image format that doesn't contain an alpha channel, then convert them back from that intermediate format to TIFF. I stick a post-it note onto the blank page for the camera auto-focus look at.
Does your camera support focus lock? If it does, you could set the focus once and it would remain set for shots of subsequent pages.

These will need holes in the glass to attach the hinge making it more expensive because of the holes. The glass door pivot hinge that I linked to in my previous reply doesn't require you to drill holes in the glass to attach the hinge. Have you seen images produced by Hiroshi's scanner? From the location of the light source in his design it will be visible as a reflection off of the platen glass. Can go at 900-1500 pages per min (see video) which is almost as fast as many 2 camera scanners and faster than some. Speed depends on skill, type of trigger and size of book.
